Hi all,
I'm interested in imaging some marine invertebrate specimens using a table-top SEM (e.g., JEOL JCM-6000, Phenom Pure, Hitachi TM3030) without sputter coating at 5 kV and then extracting DNA. The DNA will be used at template for PCR amplification of
mitochondrial and nuclear rRNA fragments. Does anyone have consistent experience with this sort of thing working well? Or do I need to invest in a cooling stage?
