OTN frame has overhead bites and payload bytes. I have read that
frame transmission happens byte wise from left to right and then
top to bottom .
Can someone please let me know the sequence.
Is it that row 1 bytes 1 to 4080 is transmitted first and then
row 2 bytes?
Or first all 4 rows of overhead is transmitted from
left, followed by payload?

This is correct:
per row 16 bytes overhead, 3808 bytes payload, 256 bytes FEC.
Or first all 4 rows of overhead is transmitted from
left, followed by payload?
This is incorrect.
It is all described in the standards: ITU-T Recommendation G.709.
Especially figure 11-2.
